Title: High Resolution of Whole-Mount Mouse Organs with Axially Swept Light Sheet Microscopy
Abstract: Medical imaging plays an integral role in clinical diagnosis or pursuing basic research. With the use of light-sheet microscopy, in combination with tissue clearing and staining techniques, researchers have been able to view high-resolution images of tissues on the subcellular level. Current light-sheet fluorescent microscope (LSFM) is associated with many advantages since LSFM has two different objective lenses for detection and illumination. Therefore, unlike other microscopy systems, field-of-view is not depending on detection objective lens magnification which allows large-of-view. However, due to tradeoff of lateral resolution and Rayleigh length (depth of focus), there is a certain limitation of mesoscale to macroscale sample imaging. In this project, undergraduate students seek to customize and produce quantifiable improvements to light sheet fluorescence microscopy (LSFM) technology and current tissue clearing and staining techniques. Customizing the LSFM will include a Rayleigh length and enhancement of nanometer spatial resolution by using Axially Sweeping Sub Voxel Light Sheet Microscopy (AS-SLSM). Furthermore, dyes and antibodies will be studied to identify optimal tissue visualization and labeling results. Finally, improved tissue clearing protocols of previously established techniques will be explored to homogenize the optical properties of the tissues.
