Title: Assessing the Long-term Degradation Behavior of Injectable Bioadhesive
Abstract: Bioadhesive has gained popularity in a wide variety of clinical applications, such as tissue adhesive, hemostats, and tissue sealants. Most recently, growth factor-releasing bioadhesive has been shown to promote the regeneration of poor-healing tissue. For that, our lab has developed injectable click chemistry-based chitosan bioadhesive for repairing shoulder soft tissue following joint injury. This study investigates the in vivo degradation of injectable bioadhesive, since the degradation behavior of adhesive within biological tissue is crucial for evaluating its efficacy and biocompatibility. For that, 50 uL of the bioadhesive was injected into the subdermal pocket of the rat skin. After implantation for various periods of time (day 0, day 7, and day 28), the tissue was then isolated for histological analysis. Hematoxylin and eosin (H&E) staining will be used to visualize the bioadhesive within the tissue. Using microscopic images and ImageJ analysis, the bioadhesive area will be calculated within the skin tissue. The degradation rate will be assessed by quantifying the percentage area of the remaining bioadhesive area over time. This study will provide insight into the bioadhesive's stability and degradation profile, which is essential for optimizing its formulation for clinical applications.
